摘  要:采用单因素实验对Enterobacter sp.SYA2产乳糖酶条件进行优化,优化后的发酵条件为:培养温度37℃、装液量50mL/250mL、摇床转速175r/min、接种量5%(V/V)、种龄8h,此条件下酶活为9.02U/mL;通过Plackett-Burman设计和Box-Behnken设计对菌株发酵培养基进行了优化,得到的最佳发酵培养基配方为:乳糖31.1g/L、蛋白胨26.0g/L、酵母膏5.0g/L、K2HPO43.0g/L、NaCl6.0g/L、MnCl20.5g/L、pH6.5,优化后的酶活为15.95U/mL。Firstly,fermentation conditions for lactase by Enterobacter sp.SYA2 were optimized by single factor experiment,the result showed that the optimum temperature was 37℃,liquid volume in flask was 50mL /250mL, vorotation speed was 175r /min,inoculation volume was 5% ( V/V) ,seed age was 8 hours.Then Plackett- Burman design and Box-Behnken Design were combined to optimize the medium components of lactase production.The results showed that the optimal medium were as follows:lactose 31.1g /L,peptone 26.0g /L,yeast extract was 5.0g /L, K 2 HPO 4 3.0g /L,NaCl 6.0g /L,MnCl 2 0.5g /L,initiation pH was 6.5. After optimization,the lipase activity was 15.95U /mL.
