超高效液相色谱法测定食品中5种天然儿茶素

摘  要:目的建立4类食品中5种天然抗氧化剂儿茶素类化合物(GCG、EC、ECG、EGC和EGCG)的超高效液相色谱-二极管阵列检测方法(UPLC-PDA)。方法样品经液液萃取、离心和浓缩后,用超高效液相色谱分离检测。结果本法对5种儿茶素均能在11min内很好分离,检测限仅0.2~2.5μg/g;空白样品的加标回收率77.7%~113.2%,相对标准差为1.1%~8.1%,峰面积在0.1~200μg/mL,呈良好的线性关系,决定系数R2均大于0.99。结论超高效液相色谱法分离速度快,且检测的灵敏度高于传统方法。Objective To establish a method for detecting 5 kinds of natural phenolic antioxidants catechins(GCG,EC,ECG,EGC and EGCG) from 4 kinds of foods by UPLC-PDA.Methods The analytes were extracted from the foods,followed by centrifugation,concentration and detection by UPLC.Results The five kinds catechin and the levels were identified and determined by.UPLC-PDA in 11 minutes,the detection limits were 0.2-2.5 μg/g only.The recovery rates of catechins,spiked blank samples were ranged from 77.7% to 113.2%,and relative standard deviations were 1.1%-8.1%.There was a good linear corrlation between the peak area and correlation of caichins with the peak area 0.1-200 μg/ml,the coefficient of determination(R2) was 0.99 or more.Conclusion UPLC method shows high separation speed and high sensitivity,comparing to common method.
