摘 要:采用001×7、D001、D301吸附树脂对酵母抽提液中的谷胱甘肽进行分离,分别考察了洗脱液(氨水、氨性乙醇、盐酸及氯化钠)浓度,离子交换柱(Φ2.5cm×30cm)装柱高度,上样流速和洗脱流速对谷胱甘肽吸附和洗脱效果的影响。结果表明,D301树脂对谷胱甘肽的吸附分离效果最好,吸附率达到84.63%;60%vol乙醇+10%氨水作为洗脱剂的洗脱效果较好,达85.51%;D301树脂装柱高度为20cm,上样流速和洗脱流速分别为1mL/min和2mL/min,此时,谷胱甘肽分离得率达到62.3%。Three adsorption resins, including 001 x7, D001, D301 were used to separate glutathione from the extract of yeast cells. The effects of elu- ant (ammonia, ammonia in ethanol, hydrochloric acid and sodium chloride) concentrations, column height of ion exchange column (qb2.5cmx30cm), sample flow rate and elution flow rate on the adsorption and elution of glutathione were investigated, respectively. The results showed that D301 resin had relatively high absorption capacity of glutathione, the absorption rate of which reached 84.63%. Using a combination of 60%vol ethanol and 10% ammonia as the eluant achieved an elution effect as high as 85.51%. When the column height of D301 resin was 20cm, sample flow rate 1.0ml/min, elution flow rate 2.0ml/min, the yield rate of glutathione separation reached 62.3%.
