摘 要:目的通过大蒜素和蒜氨酸+蒜氨酸酶混合物处理高血脂小鼠,比较了它们对高血脂小鼠体重、肝重、肝脏细胞形态和脂肪变性的影响.方法大蒜素与蒜氨酸+蒜氨酸酶混合物处理高血脂小鼠,8周后称量小鼠体重和肝重.制作小鼠肝脏细胞石蜡切片和冰冻切片观察的细胞形态.结果大蒜活性物质大蒜素与蒜氨酸+蒜氨酸酶混合物能抑制高血脂小鼠体重和肝重的增加,降低肝系数,并且具有剂量依赖性.组织学观察表明高剂量大蒜素与蒜氨酸+蒜氨酸酶混合物均能减轻脂质对肝细胞形态的改变.与高血脂模型组相比,高剂量大蒜素与蒜氨酸+蒜氨酸酶混合物处理组肝索排列呈放射状,肝细胞排列较为紧密、规则,细胞核位置接近中央,有细胞界限.细胞内颗粒、脂滴和炎性细胞浸润和肝细胞坏死较少.结论大蒜素和蒜氨酸+蒜氨酸酶混合物能减轻高血脂小鼠脂肪对肝细胞的浸润和脂肪变性,并减轻脂肪对肝小叶和细胞形态的改变.蒜氨酸+蒜氨酸酶混合物在降低高血脂小鼠体重和肝重的效果优于大蒜素.Objective The effects of allicin and mixture of alliin and allinase on the liver and body weight, hepatocyte morphology and steatosis were compared by allicin and mixture of alliinase and alliin treated hyperlipidemia mouse. Methods The liver and body weight were weighed respectively after allicin and mixture of Mliinase and alliin treated hyperlipidemia mouse for eight weeks. The paraffin and frozen section of liver cell were made to observe cell morphology. Results Both the allicin and mixture of alliin and allinase can inhibit the increase of body and liver weight in hyperlipidemia mouse and reduce liver coefficient, and they all exhibited dose-dependent. The observation of histology indicated that both allicin and mixture of alliin and allinase can alleviate the cellular morphology change by lipid induced. Compared with hyperlipidemia model group, the groups treated with high doses of allicin and mixture of alliin and allinase were found that the hepatic cord radiated out in all directions, hepatocytes arranged more closely and rules, and had cell boundary, the nuclei located in cellular centre. Furthermore, intracellular granules, lipid droplets, inflammatory cell infiltration and liver cell necrosis were less by compare hyperlipidemia model group. Conclusion Allicin and mixture of alliin and allinase can reduce the effect of lipid on hepatocyte infiltration, steatosis, and reduce changes of hepatic lobule and cell morphology in hyperlipidemia mouse. The mixture of alliin and allinase is superior to allicin on reducing liver and body weight.
