摘  要:从预制备豆制品的大豆粉浆中筛选菌株,经酪蛋白初筛及大豆分离蛋白复筛并进行SDS-PAGE验证后,得到了一株能够高效降解β-伴大豆球蛋白α亚基、α'亚基和部分β亚基的Bacillus sp.BBE201108菌株。通过单因素碳源、氮源、无机盐及正交实验确定了Bacillus sp.BBE201108的最佳培养基配方为:葡萄糖2%、大豆蛋白胨2%、酵母粉1%、Na2HPO4·12H2O0.4%、KH2PO40.06%,同时,还研究了起始pH值、培养温度对菌株产酶的影响。在最佳培养条件下,蛋白酶活力提高了3倍,最高可达292.74U/mL。A strain which can remove α-subunit,α’-subunit and partial β-subunit of β-conglycinin was isolated from soybean slurry,and then the selected strain was identified as Bacillus sp.BBE201108.The screening strategy included casein plate selecting,soybean isolate protein re-screening and SDS-PAGE verification.Based on fermentation optimization of carbon resources,nitrogen resources and orthogonal experiment,the result showed that the optimum fermentation medium consisted 2% glucose,2% soybean peptone,1% yeast extract,0.4% Na2HPO4·12H2O and 0.06% KH2PO4.Moreover,the effects of primary pH and temperature on protease production were also studied.Under the optimum conditions,protease activity was improved by 3 times,up to a maximum of 292.74 U/mL.
