摘  要:通过静态、动态相结合的方法,以葡萄多酚类物质吸附率、吸附量和解吸率为指标,确定最佳树脂型号和主要参数。结果表明,ME-1型树脂具有最佳的吸附洗脱参数,其最佳参数:在葡萄多酚含量为17.335mg/g,吸附流速为2mL/min条件下,树脂静态吸附平衡时间为10h,动态吸附最大上样量为400mL,动态洗脱多糖和蛋白杂质蒸馏水用量分别为150mL和1 300mL,乙醇洗脱剂体积分数为75%,用量为100mL时洗脱效果最好,葡萄多酚类物质的回收率可达84.8%。可见,ME-1能够较好的分离纯化葡萄枝条中的多酚类物质,且操作简单、安全。An optimal macroporous resin and its primary adsorption-desorption parameters for purification of grape polyphenols separated from grapevine cane were investigated in the present study.Static and dynamic adsorption and desorption behaviors were evaluated for the optimal macroporous and main parameters.Results showed that ME-1 resin had the excellent adsorption and desorption performance.More than 84.8% grape polyphenols were recovered under the conditions of crude extract concentration at 17.335 mg/g and a flow rate of 2 mL/min,static adsorption equilibrium time 10 h flowing through ME-1 resin column with and a maximum injection volume of 400 mL,and the saturated adsorption column washed polysaccharide and albumin impurities with 150 mL and 1 300 mL distilled water respectively and eluted with 100 mL of 75% ethanol.These results demonstrated that ME-1 resin could well purify grape polyphenols from grapevine cane and had practical value for industrialization manufacture.
