摘  要:采用液体深层发酵方式研究中药天麻对灰树花胞外多糖合成的影响,并用苯酚-硫酸法测定总糖含量。结果表明:天麻添加量为7g/L时能显著促进灰树花胞外多糖产量的增加。此时灰树花产胞外多糖产量平均为3.91g/L,与对照组的3.72g/L相比提高了5.1%。同时,对灰树花多糖合成代谢途径中的两个重要酶的酶活力进行测定。说明添加天麻成分发酵后,磷酸葡萄糖异构酶酶活力有所降低,磷酸葡萄糖变位酶酶活力则无明显变化。The effect of Rhizoma Gastrodiae on the biosynthesis of expolysaccharide (EPS) from Grifolafrondosa in submerged culture was studied. Total polysaccharide content was determined by phenol-sulphuric acid method. The results showed that Rhizoma Gastrodiae at 7 g/L significantly promoted the biosynthesis of EPS in Grifolafrondosa when compared with blank control, increasing EPS yield from 3.72 g/L to 3.91 g/L. The measured activities of two key enzymes involved in the synthesis of EPS indicated that after fermentation in the presence of Rhizoma Gastrodiae, PGI activity was decreased, although a-PGM activity showed no obvious change.
