摘  要:为研究珍稀食用菌大杯伞胞外多糖的抗氧化活性,采用菌丝液体发酵、乙醇沉淀获得胞外粗多糖,并利用DEAE 52-纤维素和Sephadex S-200凝胶柱层析对胞外多糖进行分离、纯化,以及抗氧化活性鉴定,结果表明:从胞外多糖分离到一种分子量均一的多糖CEP II,其抗氧化活性随着多糖质量浓度的增加而增加,清除自由基.OH,DPPH和.O2-的EC50值分别为0.119,0.185,0.215g/L;CEP II抗氧化活性显著,可有效去除羟基自由基、DPPH自由基、超氧自由基.In order to study the antioxidant activity of exopolysaccharides extracted from the rare edible mushroom species Clitocybe maxima, the crude exopolysaccharides were collected by submerged fermen- tation and ethanol precipitation, deproteinized with the Sevag method, and purified by DEAE 52-cellulose and Sephadex S-200 column chromatography, and then the antioxidant activity was tested. Only one com- ponent, named CEP II, was obtained from the exopolysaccharides, and its antioxidant activity was in a concentration-dependent way. Its EC0 values for scavenging OH, DPPH and O2 were 0. 119 g/L, 0. 185 g/L and 0. 215 g/L, respectively. The purified polysaccharides in C. maxima showed a strong antioxidant activity and could effectively remove free radicals, DPPH radicals and superoxide radicals.
