摘  要:以胆盐水解酶的活性为响应值,采用响应面设计分析法对植物乳杆菌发酵产胆盐水解酶培养基的成分进行优化。获得最佳培养基组成(g/L):葡萄糖19.40、蛋白胨17.05、大豆蛋白胨14.00、乙酸钠1.82、K2HPO42.00、酵母浸粉6.00、柠檬酸氢二铵1.00、Mg-SO40.87、MnSO40.45、L-半胱氨酸0.5、吐温-80 2.2mL/L。在该优化条件下,胆盐水解酶产量为(7.02±0.28)U/mL,较单因素优化前提高了6.44倍。所得数学模型的预测值与实验观察值相符,能够预测不同培养条件下植物乳杆菌发酵产胆盐水解酶产量的变化。Using activity of bile salt hydrolase as the response value,the medium composition for bile salt hydrolase production by Lactobacillus plantarum was optimized by response surface design.The optimized composition of medium were as follow(g/L): glucose 19.40,peptone 17.05,soy peptone 14.00,sodium acetate 1.82,K2HPO42.00,yeast extract 6.00,citric acid 1.00,diammonium hydrogen 1.00,MgSO40.87,MnSO40.45,L-cysteine 0.5,Tween-80 2.2ml/L.Under the optimal conditions,the production of bile salt hydrolase was(7.02±0.28)U/ml,increasing 6.44 times than before.The value predicted by the mathematical model was consistent with the experimental observations,which could be used to predict the change of L.plantarum fermentation for bile salt hydrolase production under different medium composition.
