摘  要:为了优化超声辅助提取喜树果实中桦木酸工艺条件,在单因素实验基础上,利用Design Expert.V8.0.6.1软件,设计了3因素、3水平的Box-Behnken中心组合实验,进行响应面法分析,研究了超声提取时间、乙醇体积分数、料液比对提取工艺的影响。结果表明,超声辅助提取喜树果实桦木酸的最佳工艺条件:提取时间为34min、料液比1∶30、乙醇体积分数76%、提取温度为30℃,此条件下,桦木酸理论提取含量为16.71mg/g,基本与实际提取含量(16.55±0.11)mg/g一致,说明模型拟合度较好,此法优化后的工艺稳定可靠。此外本研究发现喜树果实中桦木酸含量较高,这有益于进一步提高喜树果实的利用价值。To extract betulinic acid from Camptotheca acuminata fruits using ultrasonic-assisted extraction and to optimize the extraction conditions,the effects of ultrasonic-assisted extraction time,the volume of ethanol,the ratio of solid to liquid,and the temperature on extraction yield of betulinic acid were studied.Base on the single factor test,a Box-Behnken design involving three factors at three levels was generated by the Design-Expert software and the response surface methodology was used to optimize ultrasonic-assisted extraction conditions. The optimized conditions were as follows:34min of extracting time, 1:30 of the ratio of solid to liquid, and 76% of the volume of ethanol.The actual extraction yield of betulinic acid from Camptotheca acuminata fruits was( 16.55±0.11) mg/g closed to the predicted yield 16.71mg/g.The ultrasonic-assisted extraction conditions of betulinic acid from Camptotheca acuminata fruits using Box-Behnken design and response surface methodology was well established and optimized. Moreover, the results showed that the yield of betulinic acid in Camptotheca acuminata fruits was relatively high, it was beneficial to further improve the value of the Camptotheca acuminata fruits.
