摘 要:交联酶聚集体(CLEAs)是一种高活性的无载体固定化酶,常用的基于溶液法(Sheldon法)制备的CLEAs呈无定形的集簇体,影响其连续使用效率.以海藻糖合酶为模型酶,首次在油包水乳化体系中构建了CLEAs,利用扫描电镜和光学显微镜分析了所得CLEA的微观结构,考察了CLEAs的活性和重复使用性能.结果表明,乳化法制备的CLEAs外观呈较规则的微球形,粒径约20~60μm,其活性和重复使用性能均高于参照Sheldon法制备的CLEAs.Cross-linked enzyme aggregates (CLEAs) is a kind of carrier-free immobilized enzyme with high activity. However,the CLEAs prepared by solution based process (Sheldon's method) forms into amorphous clusters,affecting its efficiency of continuous utilization. A new approach to construct CLEAs in water-in-oil emulsion was proposed, and the microstructure and catalytic properties of CLEAs of trehalose synthase prepared by the above two methods were investigated. Optical and scanning electron microscopy of CLEAs revealed that the CLEAs obtained by emulsion based process formed into structured spherical particles between 20~ 60μm in diameter. Its activity and reusability were higher than that of the CLEAs prepared us- ing Sheldon's method.
