Optimization purification of proanthocyanidins from faba bean episperm

摘  要:利用有机溶剂精制法和大孔树脂法对蚕豆皮原花青素粗提物进行分离纯化,并对纯化产物中的原花青素组成进行研究。结果表明:经石油醚沉淀和丙酮分层后的纯度分别为47.2%、58.77%,选取AB-8型大孔树脂为最佳材料,最适宜上样浓度、上样速度、洗脱溶剂体积分数、洗脱速度分别为8 mg/mL、2 BV/h、70%乙醇、2 BV/h,最高洗脱率可达95.25%,其蚕豆皮原花青素纯度能达到94.33%。经质谱分析,分离出的蚕豆皮原花青素主要是聚合度在6以下的低聚体。Using organic solvent extraction and macroporous resin on the crude extracts of faba bean episperm proanthocyanidins for isolation and purification, and studied the composition of faba bean episperm proanthocyanidins. The results showed that: the purity of faba bean episperm proanthocyanidins can up to 47.2% and 58.77% after petroleum ether precipitation and acetone layered. Selected AB-8 macroporous resin for the best materials, the most suitable concentration of the sample on the kind of speed, the elution solvent volume fraction, the elution speed was 8 mg/mL, 2 BV/h, 70% ethanol, 2 BV/h, the washout rate can up to 95.25%, the procyanidins purity of the broad bean skin reached 94.33%. Analysis of the samples by ESI-MS, the faba bean episperm proanthocyanidins degree of polymerization in the following six.
