摘 要:目的:研究芦荟黄酮体内抗氧化活性及对CAT mRNA表达的影响。方法:采用超声波萃取,真空浓缩,冷冻干燥,用聚酰胺树脂纯化制备黄酮。用芦荟黄酮对高脂血症小鼠进行干预后,测定肝脏和脂肪组织T-AOC、CAT、GSH-Px、MDA的含量,以此评定其体内抗氧化功能。采用半定量PCR法测定肝脏和脂肪组织中CAT mRNA表达水平,从分子水平探讨芦荟黄酮抗氧化的机制。结果:芦荟黄酮能显著提高肝脏和脂肪组织中TAOC、CAT、GSH-Px的活性,抑制MDA的水平;同时可显著增强肝脏和脂肪组织中CAT mRNA的表达量。结论:芦荟黄酮具有显著的抗氧化和提高CAT mRNA表达水平的作用。Objective: To study the anti-oxidant activty and CAT mRNA expression of Aloe flavonoids in vivo. Methods: Flavonoids were preperaed with ultrasonic extraction, freeze-drying, vaccurn concentration and polyamide polymer purification. Then the flavonoids were feed to the hyperlipemia mice which induced with high fat emulsion for tour weeks, and the T-AOC, CAT, GSH-Px, MDA concentrations were detected in the liver and adipose to evaluate the an- ti-oxidant activity; meanwhile the CAT mRNA expressions were investigated by semi-quantitative PCR in the liver and the adipose to explore the anti-oxidant mechanism of the Aloe. Results: Aloe flavonoids could significantly enhance the T-AOC, CAT, GSH-Px activities and decrease the MDA level, and improve the CAT mRNA expression levels in the liver and adipose. Conclusion: Aloe flavonoids had good anti-oxidant effceets and improved the CAT mRNA expression in vivo.
