摘  要:采用水提醇沉法得到青钱柳多糖并测定其基本理化性质,同时,采用体外模拟人体胃肠道的消化模型,通过测定体外消化后产物的分子质量、还原糖含量和游离单糖含量,研究青钱柳多糖在体外模拟胃和胃肠道中的消化与吸收的情况。结果表明:青钱柳多糖的中性糖含量为31.17%,糖醛酸含量为38.44%,是一种果胶类的多糖;青钱柳多糖在体外模拟胃中会产生絮凝沉淀,而在体外模拟胃肠道的肠道消化阶段絮凝沉淀消失;青钱柳多糖经模拟胃和胃肠道消化后上清液和沉淀物分子质量没有发生变化,与多糖水溶液相比,多糖胃电解质溶液的高效凝胶渗透色谱的保留时间提前,同时,消化产物的还原糖和游离单糖含量也没有改变。结论:青钱柳多糖在体外模拟胃和模拟胃肠道均未被消化;但体外模拟胃中酸性环境和胃电解质溶液体系分别对青钱柳多糖的稳定性和构象产生了一定的影响。Polysaccharide was isolated from Cyclocarya paliurus leaves by water extraction and ethanol precipitation and its primary physico-chemical property was determined. Digestion and absorption of polysaccharide from Cyclocarya paliurus leaves was studied by employing a simulated vitro upper gastrointestinal model and analyzing molecular weight, the content of reducing sugar and free monosaccharide of digestion products at the different digestion time. The results indicated polysaccharide is a kind of pectic polysaccharide with the content of neutral sugar and uronic acid of 31.17% and 38.44%, respectively. And polysaccharide formed flocculation in vitro gastric model, however the flocculation eliminated in vitro intestinal digestion. Digestion productions of molecular weight at different digestion time remained unchanged in vitro simulated gastric and upper gastrointestinal model and compared with aqueous solution, the retention time of polysaccharide dissolved with the gastric electrolytes was shorter in high performance gel permeation chromatogram. Meanwhile, reducing sugar and free monosaccharide of digestion productions also kept unchanged in vitro simulated gastric and upper gastrointestinal model. Conclusion: polysaccharide was not digested in vitro model, but to some extent, stability and configuration of polysaccharide was influenced by acid environment in vitro simulated gastric model and gastric electrolytes, respectively.
