摘 要:研究海参皂苷(Sea cucumber saponin)对自发性糖尿病小鼠糖代谢的影响。通过体外实验观察海参皂苷对α-糖苷酶、α-淀粉酶等糖代谢相关酶的体外抑制活性。选用雄性C57BL/KsJ(db/db)小鼠,分为模型组(C57BL/KsJ(db/db))、皂苷组(C57BL/KsJ(db/db)),皂苷组饲料添加0.07%海参皂苷,另外的C57BL/ksJ为正常对照,连续喂养5周,观察海参皂苷对糖尿病小鼠空腹血糖、空腹胰岛素和葡萄糖耐受量的影响。结果显示,海参皂苷可明显抑制α-糖苷酶的活性,对α-淀粉酶抑制作用较弱。海参皂苷可降低自发性糖尿病小鼠的空腹血糖(p<0.01)、血清胰岛素(p<0.05)并改善其糖耐量(p<0.05)异常。提示海参皂苷具有良好的降血糖及改善胰岛素抵抗的作用,其降血糖机制可能与对α-糖苷酶、α-淀粉酶等糖代谢相关酶的活性抑制有关。This study was aimed to evaluate the antidiabetic effect of sea cucumber saponin on the spontaneous diabetes mice. Inhibition of the sea cucumber saponin on alpha glycosidase,alpha amylase,which were related with the glucose metabolism enzymes were observed in vitro experiment. Male C57BL/KsJ(db/db) and C57BL/KsJ were equally divided into three groups, normal group (C57BL/KsJ), model group (C57BL/KsJ (db/db) and sea cucumber saponin group(C57BL/KsJ(db/db). Mice in Saponin groups feed add 0.07% sea cucumber saponin feed for five weeks. Fasting plasma glucose level ,fasting insulin level and glucose tolerance of all groups were determinded. Sea cucumber saponin could obviously inhibits alpha glycosidase activity,certain inhibition to alpha amylase also. Sea cucumber saponin group could reduce the fasting blood glucose(p〈0.01),serum insulin(p〈0.05) and improve the glucose tolerance abnormal (p〈0.05) of spontaneous diabetes mice. The results suggested that sea cucumber saponin had good effect on controlling blood glucose level and improving insulin resistance of spontaneous diabetes mice,its hypoglycemic mechanism might be related to the inhibition on glucose metabolism enzymes.
