摘  要:研究斜卧青霉(Penicillium decumbens)产β-半乳糖苷酶的发酵条件。在单因素实验的基础上,设计正交试验,依据极差分析确定最优产酶条件。结果表明:最佳产酶条件下(乳糖70 g/L,蛋白胨10 g/L,酵母粉10 g/L,K2HPO42.5 g/L,发酵温度30℃,发酵初始pH 6.0,接种量106个/mL,装液量50 mL/250 mL,于180 r/min摇瓶培养32 h),菌体产生的β-半乳糖苷酶酶活可达153.27 U/g,是优化前的5倍。在此发酵条件下,通过高效液相色谱分析发酵液中的低聚半乳糖产量为30.2 g/L。The fermentation conditions of β-galactosidase produced from Penicillium decumbens were re- searched. The orthogonal experiment was designed based on the single factor experiments. The optimal fer- mentation conditions were ascertained by range analysis. The results showed that the optimal fermentation conditions for β- galactosidase production were lactose concentration 70 g/L, peptone 10 g/L, yeast 10 g/L, K2 HPO4 2.5 g/L, fermentation temperature 30 ℃, the initial pH value 6.0, inoculum size 106/mL, liquid volume 50 mL/250 mL, rotation speed 180 r/min and fermentation time 32 h. Under these condi- tions the activity of 13 - galactosidase would reach 153.27 U/g which was five times of that before the op- timization. The yield of galaetooligosaceharide in the fermentation broth was 30.2 g/L analyzed by HPLC.
