摘 要:目的研究海参由来的脑苷脂及神经酰胺对酵母浸粉诱导高尿酸血症的改善作用及其机制。方法通过喂食雄性昆明小鼠20%的酵母浸粉建立高尿酸血症模型,比较了摄食整参及其所含有的脑苷脂(0.02%,0.04%)对血尿酸的影响。在此基础上,利用相同的实验动物模型,比较了海参由来的脑苷脂及神经酰胺对血尿酸的影响,并分别测定小鼠肝脏黄嘌呤氧化酶(XOD)和腺苷脱氨酶(ADA)活力及其mRNA表达量,对脑苷脂及神经酰胺的降血尿酸作用机制进行了初步探讨。结果摄食海参及其活性成分脑苷脂均可显著抑制酵母浸粉诱发的血清尿酸水平升高,且0.02%的低剂量与0.04%的高剂量效果相同。海参由来的神经酰胺对血清尿酸水平也有显著的降低作用。同时,脑苷脂及神经酰胺均可显著抑制小鼠肝脏XOD及ADA酶活力和mRNA表达量,XOD酶活力分别降低了44.9%及34.8%,ADA酶活力分别降低了25.5%及20.7%;XOD mRNA表达量分别降低了44.6%及31.5%,ADA mRNA表达量分别降低了44.1%及38.7%。结论海参脑苷脂及其主要结构单元神经酰胺对酵母浸粉诱导的高尿酸血症有明显改善作用,其机制与降低了肝脏XOD mRNA和ADA mRNA表达量从而抑制相关尿酸生成酶活力有关。Objective To study the effects and mechanisms of cerebroside and eeramide from sea cucumber on hyperuricemia in mice fed with high yeast powder. Methods Male KM mice fed with 20 % yeast pow-der were used to establish the hyperuricemia model. We compared the effects of whole sea cumber and its cerebroside (0. 02%, 0.04%) on serum uric acid. On the basis of that, we compared the effects of cerehroside and ceramide from sea cucumber on serum uric acid by using the same experimental animal model. In addition, xanthine oxidase (XOD) and adenosine deaminase (ADA) activity and mRNA ex-pression levels in mice liver were determined for the mechanism preliminarily. Results Whole sea cu-cumber body, eerebroside and ceramide all decreased the serum uric acid level significantly. Both cere-broside and ceramide could significantly inhibit XOD and ADA enzyme activity and mRNA expression levels in mice liver. In detail, XOD enzyme activity was reduced by 44. 9% and 34. 8% respectively, ADA enzyme activity was reduced by 25.5% and 25.5% respectively; XOD mRNA expression level was reduced by 44. 6G and 44. 6% respectively, ADA rnRNA expression level was reduced by 44. 1% and 44. 1% respectively. Conclusion Cerebroside from sea cucumber and its main structural unit ceram-ide have improvement on hyperuricemia induced by high yeast powder in mice.
