摘  要:本研究采用体外实验,分离鉴定人正常食管上皮细胞,用RT-PCR方法测定伏马菌素B1(fumonisin B1,FB1)对细胞周期相关基因Cyclin D1、Cyclin E、P16、P21、P27mRNA表达的影响。结果表明:FB1作用人正常食管上皮细胞后可以在转录水平上影响Cyclin D1、Cyclin E、P16、P21、P27基因的表达,导致Cyclin D1mRNA高表达,Cyclin E、P16、P21、P27基因mRNA低表达。提示,细胞周期相关基因的异常表达与癌症密切相关,该研究从体外实验进一步证实FB1可能与人类食管癌的发生有关。Epidemiological studies have shown that fumonisin Bt (FB1), a fungal toxin, may be associated with esophageal cancer, but the laboratory evidence is still limited. In this study, human normal esophageal epithelial cells were isolated and identified in vitro. The influence of FB1 on the mRNA expression of cell cycle-related genes including Cyclin D1, Cyclin E, P16, P21, and P27 was determined by reverse transcription-polymerase chain reaction (RT-PCR). The results showed that FB1 played an important role in regulating the expression of Cyclin D1, Cyclin E, P16, P21, and P27 genes in normal human esophageal epithelial ceils at the transcriptional level, leading to high mRNA expression of Cyclin D1 but low mRNA expression of Cyclin E, P16, P21, and P27 genes. The abnormal expression of cell cycle-related genes is closely related with cancer. This in vitro study has further confirmed that FB1 is related with the occurrence of human esophageal cancer.
