摘  要:采用超微粉碎技术处理原料玉米蛋白粉,在Plackett-Burman设计、最陡爬坡试验的基础上选取加酶量、微粉粒度、微波功率为自变量,并以玉米黄色素的提取率为响应值进行响应面优化设计。研究结果表明:玉米黄色素最大吸收峰为447 nm,最陡爬坡试验表明响应中心为:加酶量1%、微粉粒度300目、微波功率300 W,最后利用Box-Behnken响应面分析建立了3个关键影响因素的三元二次多项式数学模型:Y=0.57+0.026X1+0.046X2-0.015X3+8.25×10-3X1X2-8.00×10-3X1X3+5.00×10-4X2X3-2.71×10-3X12+8.29×10-3X22+9.04×10-3X32,得到最佳提取工艺为:加酶量1.25%,微粉粒度400目,微波功率200 W。模型预测结果提取率为0.688 mg/g,验证试验结果提取率达0.672 mg/g,与预测值吻合相对良好。Raw material was treated by ultrafine comminution. Based on Plackett-Burman design and steepest ascent experiment, enzyme addition, particle size and microwave power were selected as independent variables. The extraction rate was also selected as response value to conduct the optimization of response surface design. Results showed that the maximum absorption peak was on 447 nm. Results of the steepest ascent experiment indicated that the response center was as follows: enzyme addition of 1%, particle size of 300 mesh, and microwave power of 300 W. With Box-Behnken response surface analysis, a 3-factor and 2-degree mathematical model was established as follows: Y=0.57+0.026X 1 +0.046X 2- 0.015X 3 +8.25×10-3X 1 X 2-8.00×10-3X 1 X 3 +5.00×10- 4X 2 X 3-2.71×10-3X 1 2+8.29×10-3X 2 2+9.04×10-3X 3 2. Theoptimum extraction conditions were as follows: enzyme addition of 1.25%, particle size of 400 mesh, and microwave power of 200 W. Based on the model, the extraction rated was expected to reach 0.688 mg/ g. Results of the demonstration experiment showed that the extraction rate was as high as 0.672 mg/g, in good agreement with the predicted results.
