基于单细胞凝胶电泳技术快速检测食品添加剂对V_(79)细胞DNA损伤作用的研究

摘  要:目的快速检测啤酒花浸膏、溴酸钾和焦磷酸钠等3种食品添加剂对V79细胞DNA的损伤作用。方法采用V79细胞培养和单细胞凝胶电泳技术,借鉴单细胞凝胶电泳技术(SCGE)图像分析系统IMI1.0对彗星图像进行分析,观察指标有尾长、尾重心、olive尾矩、尾惯量、尾分布矩、尾长/头长比、尾DNA含量(%)等7个计量指标。结果实验结果表明,溴酸钾和焦磷酸钠的剂量分别在75~300μg/ml和25~100μmol/ml范围内,均对V79细胞DNA有明显的损伤作用,存在剂量-反应关系;啤酒花浸膏的剂量在25~100μg/ml范围内,不引起V79细胞DNA的损伤作用。结论本研究方法与传统的彗星实验相比,具有分析指标多,操作方便等优点,可应用于快速检测食品添加剂诱变性的筛选试验。Objective To study rapid test method of the damnification for the DNA of V79 cell by European hops extract, potassium bromate and sodium pyrophosphate. Methods The single cell gel electrophoresis assay and V79 cell culture technic were used, the SCGE Image analyse system IM11.0 was adopted to analyze comet image for reference. The index included length, barycenter,square, inertia, moment of distribution, ratio of length/head length and content of DNA (%). Results The result of the study show that : It' 11 cause distinctness damnification for the DNA of V79 cell when the dosage of potassium bromate and sodium pyrophosphate were in the range of 75-300 p^g/ml and 25-100 Ixmol/ ml respectively, and it exist the relationship of dose-reaction. Meanwhile, when the European hops extract was in the range of 25-100 ug/ml, it' ll not cause damnification for DNA of V79 cell. Conclusion With the traditional comet experiment,the study has the excellence such as more analyse index, convenient operate as well.The study could apply to the filtration experiment for the rapid test of themutagenicity of food additives.
