摘  要:以罗非鱼鱼皮为原料制备鱼皮胶原蛋白,利用正交试验确定碱性蛋白酶和中性蛋白酶的最适水解条件,并进一步确定双酶复合水解最佳条件.结果表明,中性蛋白酶和碱性蛋白酶连续水解方式制备的胶原蛋白水解液具有最高的水解度和羟自由基清除能力.将该水解液依次通过Sephadex G-25凝胶色谱柱、SP Sephadex C-25阳离子交换色谱柱、Sephadex G-15凝胶色谱柱和反向高效液相色谱(RP-HPLC)进行分离纯化,得到了一种氨基酸序列为Glu-Gly-Leu的抗氧化肽,其分子量为317.33 Da,该肽对羟自由基清除能力的IC50值为4.61μg/mL.本文为罗非鱼鱼皮胶原蛋白制备抗氧化肽提供了科学依据.Tilapia skin gelatin (TSG) is obtained from the raw material of Tilapia skin. The optimum hydrolysis condition of neutrase and alcalase is then determined using the orthogonal experiment, and double-enzyme hydrolysis is further applied to obtain the best hydrolysis condition for the preparation of gelatin hydrolysate. The results show that the gelatin hydrolysate obtained by progressive hydrolysis using neutrase and alcalase has the highest DH and hydroxyl radical scavenging activity. This hydrolysate is purified thereafter by using Sephadex G -25 gel filtration chromatography, SP Sephadex C -25 ion exchange chromatography, Sephadex G - 15 gel filtration chromatography and RP - HPLC. An antioxidant peptide is obtained. Its amino acid sequences are identified as Glu - Gly - Leu, and the molecular weight is 317. 33 Da. The IC50 value of this peptide on hydroxyl radical scavenging activities is 4. 61 ug/mL. This study provides a scientific basis for the preparation of antioxidant peptides from the hydrolysate of TSG.
