摘  要:为探索啤酒生产废水的资源化利用,采用单因素和正交试验设计对斯达油脂酵母(Lipomyces starkeyi1809)利用啤酒生产废水产油脂和化学需氧量(COD)降解率的发酵条件进行优化,并采用8 L的发酵罐进行放大试验和发酵动力学分析,采用气相色谱法分析菌体油脂脂肪酸的组成。试验结果表明:最佳发酵条件为接种量3%、装液量130 mL、温度26℃、转速160 r/min、发酵时间144 h。在此条件下,菌体生物量、油脂产量、油脂含量、COD降解率和油脂不饱和脂肪酸指数(IUFA)分别达到13.83 g/L、5.25 g/L、37.92%、79.08%和65.46%,较优化前分别提高了12.62%,19.32%,5.92%,57.15%和2.36%,优化效果显著。发酵罐放大试验培养到120 h时,菌体生物量、油脂产量、油脂含量和COD降解率均达到最大值,分别为13.34 g/L、4.82 g/L、36.13%和85.09%,菌体生物量、油脂产量和油脂含量与正交试验的最优水平组合条件下所得结果相似,其中COD降解率提高了7.59%,且发酵周期缩短了1 d。发酵动力学分析表明:COD降解与菌体生长同步,属于与生长相关型,而油脂积累则属于与生长部分相关型。For achieving the reutilization of waste water resources, this study used optimized brewery products waste water to cultivate Lipomyces starkeyi 1809, the optimum conditions for lipid production and the degradation of COD was studied by single-factor and orthogonal design experiments, and using a 8 L fermentor to amplify, and conducting the fermentation dynamics analysis, and using gas chromatography analysis the fatty acid of microbes lipid. The results showed that the best conditions consisting of inoculation volume 3%, volume 130 mL/500 mL, temperature 26 ℃, shaking speed 160 r/min, culture time 144 h. Under these conditions, the biomass, the lipid production ,the lipid content, the proportion of degradation of waste water's COD and the IUFA were up to 13.83 g/L, 5.25g/L, 37.92%, 79.08% and 65.46% respectively, increased by 12.62% ,19.32%,5.92% ,57.15% and 2.36% respectively, the optimization effect was significant. When it reached 120 h of the fermentor training, the biomass, the lipid production, the lipid content and the proportion of degradation of waste water's COD all reached the maximum, they were 13.34 g/L, 4.82 g/L, 36.13% and 85.09% respectively, the biomass, the lipid production and the lipid content were similar with the result of the orthogo- nal design experiments under the best conditions, but the proportion of degradation of waste water's COD increased 7. 59%, and the fermentation cycle abridged one day. The fermentation dynamics analysis showed that the COD's degrada- tion keeping pace with the thalli's growth and belonging to the type which was related with the growth, and the lipid's accumulation belonging to the type which was related with the growth partially.
