摘  要:采用响应面法优化杜仲叶中总多酚超声波辅助提取工艺。考察了提取溶剂、液料比、提取时间、提取温度及提取次数对提取工艺的影响,在单因素实验分析的基础上采用Box-Benhnken中心组合进行4因素3水平的实验设计,以总多酚得率为响应值,进行响应面分析,建立二次多项回归数学模型,并优化提取工艺。结果表明杜仲叶中总多酚超声波辅助提取最佳工艺为:55%乙醇、液料比为25∶1mL·g-1、45℃下提取25min,提取两次,在此条件下,总多酚得率为4.678%。Response surface analysis methodology( RSM) was used to optimizing the ultrasonic assisted extraction technology of total polyphenols in leaves of Eucommia ulmoides Olive. The influence on the technology caused by the variety of extraction solvents and the concentrations,liquid-to-solid ratio,extraction time,extraction temperature and extraction times was investigated. Experiments were arranged according to Box-Behnken central composite experiment design based on results of single-factor experiments. Response surface analysis was applied to create the two multinomial mathematical and optimize the extract process.Results showed that the optimum parameters for extracting from leaves of Eucommia ulmoides Olive were 55%ethanol,25 times the amount of solvent and 25min at 45℃ twice. Under these condition,the yield of total polyphenols was 4.678%.
