摘  要:为了提高核酸提取率,增加双孢菇废弃菇柄的经济价值。以双孢菇废弃菇柄为原料,通过单因素试验研究了pH值、酶解温度、加酶量、液固比、酶解时间对核酸提取率的影响,并采用响应面法对纤维素酶法提取核酸工艺进行优化。结果表明,在单因素试验结果基础上,采用响应面分析法对主要工艺参数进行优化,得到双孢菇废弃菇柄中核酸提取最优工艺参数为:酶解温度46.5℃、pH5.4、加酶量0.06%、液固比(mL/g)26:1,理论核酸提取率3.04 g/100 g。通过验证试验,实际核酸提取率可达2.87 g/100 g,试验证明模型拟合程度良好。To improve the yield of nucleic acid, increase the economic value of Agaricus bisporus waste stipe. Nucleic acid were extracted from Agaricus bisporus waste stipe by cellulase extraction. The effects of pH, temperature, papain amount, ratio of liquid to solid, enzyme time on the yield of nucleic acid were investigated by single factor experiments, and optimized the parameters by response surface analysis. The results showed that on the basis of single factor experiments, the parameters of cellulase extraction optimized by response surface analysis are as follows: temperature 46.5 %:, pH5.4, papain amount 0.06%, liquid-solid ratio 26:1 mL/g. Under these optimum conditions, the yield of nucleic acid extract from Agaricus bisporus waste stipe can be up to 2.87 g/100 g of which slightly lower than the theoretical yield (3.04 g/100 g). This results indicate that the regression model could predict the yield of nucleic acid of from Agaricus bisporus waste stipe well.
