摘  要:为筛选能够产生稳定性较高木聚糖酶的菌株,采用以桦木木聚糖为唯一碳源的培养基,从牛瘤胃液中分离到1株产木聚糖酶的细菌,命名为BF。将BF菌株进行形态学分析、生理生化鉴定及16SrDNA鉴定,并研究其产木聚糖酶的最适反应温度、最适pH值、温度稳定性、pH值稳定性及金属离子对其酶活力的影响。结果显示,BF菌株属于类芽孢杆菌属(Paenibacillus);BF菌株所产木聚糖酶反应最适温度为60℃,最适pH值为6.0,在pH值3.0~9.0稳定,在60℃保温180min后仍有65%的酶活力,具有较好的热稳定性;终浓度为5mmol/L的ca2+、Fe2+对酶活力有较强的促进作用,相对酶活力分别达130%和140%。表明,筛选到的菌株所产木聚糖酶具有较好的稳定性。In order to obtain a new xylanase with desirable properties,the bovine rumen fluid was collected to screen bacterial strains producing xylanase by using the selective medium with birch- wood xylan as the sole carbon source. A strain named BF which could produce xylanase was isola- ted and identified to the genus of Paenibacillus based on its morphological, physiological, and bio- chemical characteristics,as well as 16S rDNA phylogenetic analysis. The optimal pH of BF strain xylanase was 6. 0 and its optimal temperature was 60 ~C. The xylanase was stable in the wide range of pH value of 3.0-9.0 and could retained 65% relative activity after incubated for 180 min at 60 ℃, showing its good thermostability. Both Ca2+ and Fez+ with final concentration of 5 mmol/L had a strong promoting effect on xylanase activity,with relative activity of 130% and 140% ,respectively.
