牛、羊初乳中多肽的筛分比较及抗炎作用研究

摘　　要：应用AKTA purifier 10蛋白质层析仪,对经过模拟人体消化后的牛、羊初乳中的多肽进行分离纯化,并比较其差异性,同时应用小鼠肿瘤坏死因子TNF-αELISA试剂盒对所分离出的多肽进行抗炎作用检测并比较,为抗炎功能性食品的开发提供参考。结果:①从牛初乳中分离出了23种多肽,从羊初乳中分离出了36种多肽;②羊初乳中分离出的多肽种类多于牛初乳;③通过检测,所有乳样品中小鼠肿瘤坏死因子TNF-α的浓度范围是(2.42±0.006)^(18.03±0.102)ng/L,与阳性对照(18.52±0.027)ng/L相比,所有分离出的多肽均具有抗炎作用;④通过比较,羊初乳第三天中含有的一种多肽显示出最好的抗炎性能。In this study, the types of peptides purified from bovine and caprine colostrums purified using an AKTA purifier 10 were deter- mined after simulating human gastrointestinal digestion. The peptide fractions obtained from the milk samples were investigated and the differ- ences were also evaluated. The anti-inflammatory activities of the peptides purified from the milk samples （AKTA fractions） were determined using competitive TNF-α ELISA. There are some bioactive peptides which have anti-inflammatory effects on human health after they digest milk, and such activities will expand their applications as functional food. The results showed that： ①Twenty-three types ofpeptides were ob- tained from the bovine colostrums and thirty-six types of peptides were obtained from the caprine colostrums. ②The caprine colostrums were demonstrated to possess more types of peptides compared to the bovine colostrums. ③ Using the competitive TNF-α ELISA method, the mean values for the concentration of TNF-① increased from 2.42±0.006 ng/L to18.03±0.102 ng/L in all of the samples. Compared with the positive control 18.52±0.027 ng/L, all of the peptides showed anti-inflammatory activities. ④Based on comparison analysis, the greatest anti- inflammatory activity of all of the samples was demonstrated for the caprine colostrums fraction. Conclusions： The caprine colostrums were demonstrated to possess more types of peptides compared to the bovine colostrums. The captine colosmum were also showed the greatest an- ti-inflammatory activity than the bovine colostrums.
