摘 要:为了解决传统玉米淀粉提取工艺时间长、污染严重等问题,以玉米为原料,采用酶与细胞渗透剂协同浸泡提取玉米淀粉,并对该工艺进行优化.通过单因素试验和响应面设计法,研究了该工艺的浸泡时间、浸泡温度、加酶量和pH值对玉米浸泡效果的影响.通过优化组合得到最佳工艺条件为浸泡时间4.54 h、浸泡温度49.74℃、加酶量17 824 U/100 g、pH值3.42,在最优条件下进行验证试验,玉米淀粉提取率为88.97%.但从实际操作和经济效益方面考虑,将数据修正为浸泡时间4.5 h、浸泡温度50℃、加酶量17 800 U/100 g、pH值3.4,此条件下玉米淀粉提取率为86.60%,实际值与理论值基本相符.表明酶与细胞渗透剂协同浸泡提取玉米淀粉的工艺是可行的.In order to solve the problems of the conventional corn starch extraction process, such as long extraction time and serious pollution, a new extraction process was developed to extract com starch by soaking with enzyme and cell penetrant. Through single factor experiments and response surface designs, the effects of soaking time, soaking temperature, enzyme dosage and pH value on the com soaking effect were studied. The optimum process conditions were determined by optimal combination as follows: soaking time 4.54 hours, soaking temperature 49.74 ~C , enzyme dosage 17 824U per 100 grams of com, and pH value 3.42. The verification test was carried out under the optimum conditions, and the corn starch yield reached 88.97%. Considering the actual operations and the economic benefits, the data were modified to soaking time 4.5 hours, soaking temperature 50 ℃, enzyme dosage 17 800U per 100 grams of com, and pH 3.4. Under the modified conditions, the corn starch yield reached 86.60%, which was basically consistent with the theoretical value. The result indicated that the synergistic soaking process with enzyme and cell penetrant was was feasible to extract corn starch.
