摘 要:目的:研究油菜蜂花粉中具有保肝功效的活性组分。方法:以四氯化碳(CCl4)诱导人正常肝细胞(L-02)建立体外细胞损伤模型,采用MTT法(噻唑兰比色分析法)测定细胞存活率,并以其作为活性筛选指标,采用溶剂萃取、反相色谱层析作为分离手段,对油菜蜂花粉各组分进行细胞学筛选,结合薄层色谱层析法(TLC)进行检测。结果:体外细胞损伤模型的造模浓度以10 mmol/L CCl4为佳,细胞活性筛选结果显示正丁醇萃取物活性最高,100μg/mL和200μg/mL剂量组细胞存活率分别为60.65%和76.98%;正丁醇萃取物各洗脱组分中A2活性最高,100μg/mL和200μg/mL剂量组细胞存活率分别为76.66%和64.47%,为油菜蜂花粉中主要的保肝活性组分。A2的定性分析及前期研究结果表明其为黄酮醇苷类物质。To investigate the hepatoprotective components in rape bee pollen,a vitro model of L-02 hepatocytes injured by CCl4was established.Cell survival rate measured by MTT assay was considered as the screening criterion,solvent extraction and reversed phase column chromatography were used as the separation method.TLC was used as a detection and analysis method.The result showed that the appropriate concentration of CCl4that could induce hepatocytes injury was determined as 10 mmol/L.N-butanol extract group showed the highest hepatoprotective effect that the cell survival rate of 100 μg/mL group and 200 μg/mL group was 60.65% and 76.98% respectively.Component A2separated from N-butanol extract showed the highest hepatoprotective effect that the cell survival rate of 100 μg/mL group and 200 μg/ mL group was 76.66% and 64.47%.The result of qualitative analysis and earlier research of A2suggested that A2may be flavonoides.
