摘  要:从本实验室保存的嗜酸乳杆菌的细胞壁中分离肽聚糖,研究其组成成分和结构。采用TritonX-100处理MRS肉汤培养物,高速离心分离不可溶物质,经三氯乙酸去磷壁酸,胰蛋白酶、核酸酶、α-糜蛋白酶消化,甲醇与氯仿除脂等步骤,分离并纯化精制肽聚糖(PG)。经多糖、蛋白质及氨基酸含量分析,所提取的物质主要是肽聚糖。经肽聚糖的溶解试验,红外光谱和高效液相色谱分析,所提取肽聚糖结构与已知肽聚糖结构相吻合,其糖链结构为D型吡喃糖N-乙酰葡糖胺和N-乙酰胞壁酸以β-(1,4)-糖苷键连接而成。最终确定提取物的主要成分为肽聚糖,经聚丙烯酰胺凝胶电泳(SDS-PAGE)分析,溶菌酶水解后的肽聚糖分子质量在60 ku左右。PG derived from Lactobacillus acidophilus saved in the laboratory was separated and purified from the cell wall, and involved in research and analysis of its chemical compositions and structure. After dealt with MRS broth cultures by Triton X-100, using high-speed centrifugal separated not soluble subs-t-ance from the MRS broth cuhuers. By trichloroacetic acid to get rid of teichoic acid;pancreatic enzyme,nucleic acid enzyme, α-chymotrypsin to digest; methanol and chloroform to Eliminate fat, separation and purification of refined peptidoglycan(PG). The major component of the preparation was validated to be peptidoglycan by animo acid, HPLC and Infrared spectrum substantiated the gly- can chain of peptidoglyc-anwas connected by D-pyranose N-acetylglucosamine and N-acetylmuramic acid through β-(1, 4)-glycosid-ic bond, SDS-PAGE and multistain suggested that the molecular weight of soluble peptidoglycan showed its 60ku.
