摘 要:目的 测定恶性疟原虫 (P f)海南株CTP基因序列 ,了解该虫株CTP基因序列与其它种类的CTP基因序列的差异 ,用于药物靶位的筛选。方法 用PCR的方法特异性的扩增CTP基因 ,并将此基因克隆于测序载体 pUC19,采用Sanger双脱氧链末端终止法测序。结果 我国海南株的CTP基因序列与其它的CTP基因有不同程度的差异。结论 P .Aim To determine the sequence of CTP gene of Plasmodium falciparum and find out the differences of the CTP gene sequence between isolate Hainan and other isolates.Methods CTP gene was amplified by polymerase chain reaction and cloned into sequencing vector pUC19.The recombinant plasmid pUC19/CTP was screened by blue-white plaque and identified by PCR and restriction analysis.The cloned CTP genes were sequenced by Sanger's method.Results There is no significant differences between P.f CTP genes of FCC1/HN and pf K1 strain,but there is obvious difference between P.f CTP genes of FCC1/HN and human CTP.Conclusion It is valuable to further study on P.f CTP as a candidate target of anti-malaria drug.
