摘 要:为选择适合于灰树花胞外多糖脱色的方法,分别采用活性炭、过氧化氢和大孔树脂D941对灰树花胞外多糖进行脱色。通过对脱色率和多糖保留率的比较发现,3种方法对灰树花胞外多糖均有脱色作用。活性炭法的脱色率为37.74%,多糖保留率为47.92%,过氧化氢法的脱色率为68.28%,多糖保留率为53.07%,大孔树脂D941脱色的脱色率为74.24%,多糖保留率为61.87%。活性炭法相比另2种方法的脱色率和多糖保留率均较低,选取过氧化氢法和树脂法脱色后的多糖进行α-葡萄糖苷酶的抑制试验,结果表明:树脂法脱色后的多糖对α-葡萄糖苷酶的抑制作用明显高于过氧化氢法脱色后的多糖(P<0.05)。用5种大孔树脂进行多糖脱色,选择脱色率和多糖保留率较高的D941和D101脱色后的多糖进行α-葡萄糖苷酶的抑制试验,结果表明:大孔弱碱性离子交换树脂D941和大孔吸附树脂D101适合用于灰树花胞外多糖脱色。In order to choose the most suitable decolorizing methods for exopolysaccharide of Grifola frondosa, the activated carbon, hydrogen peroxide and macroporous resin D941 were used to decolorize the exopolysaccharide from Grifola frondosa. Through the comparison of the retention rate and decolorization rate of polysaccharides, it was found that the three methods all had decolorization effect on the exopolysaccharides of Grifola frondosa. The decoloration rate of the activated carbon method was 37.74%, the polysaccharide retention rate was 47.92%, the decolorization rate of the hydrogen peroxide method was 68.28%, and the polysaccharide retention rate was 53.07%. The macroporous resin D941 showed a decolored polysaccharide retention rate of 61.87% and a decolorization rate of 74.24%. Compared with the other two methods, the activated carbon method has lower decolorization rate and polysaccharide retention rate. The decolorization of polysaccharides by the hydrogen peroxide method and the resin method was tested for α-glucosidase inhibition. The results showed that: The inhibitory effect of α-glucosidase was significantly higher than that of the decolored polysaccharide by the hydrogen peroxide method(P<0.05). Five macroporous resins of different types and properties were screened and found: Macroporous weak anionic resin D941 and macroporous resin D101 are more suitable for decolorization of Grifola frondosa exopolysaccharides.
