摘 要:目的为满足人们对于微量元素硒的日常膳食需求,通过将低级生物利用的无机硒代谢成高级生物可利用的有机硒,增加乳杆菌的硒含量,找出最佳培养方案。方法首次选用植物乳杆菌作为富硒载体。选用MRS液体培养基为基础培养基,探究不同接种量、碳源、氮源、pH值及发酵时间对OD值及富硒含量的影响,并对其培养条件进行优化。结果植物乳杆菌最优富硒方案为:亚硒酸钠添加量为5.0μg/mL,葡萄糖含量为20.0g/L,牛肉膏含量为24.0g/L,初始pH值为5.0,加硒时间为6h,接种量为4.0%,发酵时间为36h。优化后植物乳杆菌富硒含量达到842.118μg/g。结论通过单因素实验对富硒培养的条件进行优化,增加植物乳杆菌的硒含量,得出最佳富硒培养方案,实现将无机硒有机转化,为今后食品中安全且高效地添加有机硒元素提供可行性方案。Objective To meet people's daily dietary needs for trace element selenium,increase the selenium content of Lactobacillus and find the best culture solution by metabolizing the inorganic selenium used by low-grade organisms into advanced bio-separable organic selenium.Methods This is the first choice of lactobacillus plantamm Lb-30as seleniumenriched carrier.MRS liquid medium was used as the basic medium to explore the effects of different inoculum size,carbon source,nitrogen source,pH value and fermentation time on OD value and selenium content,and the culture conditions were optimized.Results The optimum conditions of Lb-30 were as follows:sodium sulfite concentration was 5.0μg/mL,the glucose concentration was 20.0g/L,the beef extract concentration was 24.0g/L,the initial pH was 5.0,the selenium adding time was 6h,the inoculation amount was 4.0%,the fermentation time was 36h,the final selenium content of lactobacillus plantarum reached up to 842.118μg/g.Conclusions The conditions of selenium-enriched culture were optimized by single facto/experiment,the selenium content of lactobacillus plantarum was increased,and the best selenium-enriched culture scheme was obtained,which realized the organic conversion of inorganic selenium,which is safe and efficient for future food. The addition of organic selenium provides a viable solution.
