摘  要:从干预细胞氧化应激的角度,采用比色法、荧光检测及免疫分析的方法,考察不同浓度的矢车菊-3-葡萄糖苷对环氧丙酰胺引起的人类乳癌细胞MDA-MB-231毒性的影响。试验结果表明,与环氧丙酰胺单独处理的细胞相比,矢车菊-3-葡萄糖苷预处理后能明显提高细胞存活率,显著降低细胞内活性氧和丙二醛的生成量,而且抑制细胞内还原型谷胱甘肽含量的下降,提高γ-GCS蛋白表达量。矢车菊-3-葡萄糖苷能够保护MDA-MB-231细胞免受环氧丙酰胺引起的氧化应激。Acrylamide(AA) formed in carbohydrates-enriched foods during heating processing. AA has been categorized as a possible carcinogen. Glycidamide(GA), a reactive epoxide, is the metabolite of AA in body and more toxic than AA itself. Cyanidin 3-glucoside(Cy-3-glu) is one of the natural antioxidants widely found in plants. In the present study, we investigated whether Cy-3-glu can decrease the GA-reduced cytotoxicity of human breast cancer MDA-MB-231 cells. Colorimetry assays, fluorescence detection and immunoblot analysis were performed. Results showed that Cy-3-glu significantly enhanced cell viability and reduced the generation of reactive oxygen species(ROS) and malondialdehyde(MDA) Moreover, the pretreatment with Cy-3-glu suppressed glutathione(GSH) delption and increased gamma-glutamylcysteine synthetase(γ-GCS) protein expression by GA. Cy-3-glu could present the protection against cell damage induced by GA in MDA-MB-231 cells.
