酶法破壁转化废菌体制备甲壳低聚糖

摘  要:试验采用酶法转化柠檬酸发酵后的废菌体,研究了酶法破壁及降解细胞壁几丁质制备甲壳低聚糖的工艺条件。采用红外光谱法对破壁后产物的化学结构进行表征,采用乙酰丙酮法测定几丁质酶降解产物中乙酰氨基葡萄糖还原糖的含量。结果表明,破壁产物的主要成分是(1-3)-a-D-葡聚糖和几丁质的复合物,10mg废菌丝体破壁产物中加入0.3%几丁质酶降解反应7d后可得到0.23mg乙酰氨基葡萄糖还原糖。该方法制备甲壳低聚糖条件温和、工艺简便,为发酵废渣的综合利用开辟了新途径。Waste mycelia from citric acid production plant were used to produce chi tooligosaccharides by enzymatic cell wall breaking and enzymatic degradation. The process conditions of the cell wall breaking and degradation by enzymes were investigated. FT-IR spectra were used to characterize the chemical structure of the product from the cell wall breaking. Acetylacetones method was used to determine the content of N-acetyl-glucosamine reducing sugar. The results showed that the product from the cell wall breaking was the complex polysaccharide with (1-3)-a-D-glucan and chitin. Degradation by 0.3%chitinase with 10 mg products from the cell wall breaking for 7 d, 0.23 mg of N-acetyl-glucosamine reducing sugar was obtained. This new method is devised to obtain N-acetyl-glucosamine and disaccharides with stable properties and many important biological activities and functions by mild reaction and less environmental pollution. A new way for the comprehensive utilization of fermented waste is developed.
