摘 要:分析了培养条件和营养成分包括培养温度、培养基初始pH值、培养基碳源、氮源、KH2PO4、NaCl和吐温-80对植物乳杆菌T102生长量及产细菌素能力的影响。结果表明,培养基pH值不变,30℃时的细菌素活性最高,37℃时菌体的生长量最大,在培养基初始pH值为6.0时的菌体生长量较低,但是细菌素活性要高于pH值为7.0的培养基。乳糖和酵母提取物是最佳的碳源和氮源。分别添加质量分数为2.5%乳糖、2.0%酵母提取物、2.0%的KH2PO4、1.0%的NaCl和质量浓度为1.0 g/L的吐温-80时最有利于细菌素的合成。吐温-80对菌株的生长没有促进作用,但是对细菌素的产生有很大的影响。This study was conducted to investigate the effect of cultural temperature, intial pH, carbon sources, nitrogen sources and KH2PO4,NaCl and Twain 80 on the cell growth and bacteriocin production of Lactobacillus plantarum T102. In cultures without pH control, the bacteriocin activity ofL. plantarum T102 was higher at 30℃, however, the cell growth rate was higher at 37 ℃. Ifi MRS broth with an initial pH 6.0, the cell growth was lower, but high hacteriocin levels were recorded than at pH7.0.Lactose was the best carbon sources and yeast extract was the best nitrogen sources. The bacteriocin activity was maximal in medium containing 2.5% lactose or 2.0% yeast extract or 2.0% KH2PO4 or 1.0% NaCl or 1.0 g/L Twain 80. Twain 80 didn't promote strain growth, but had great influence on bacteriocin production.
