摘  要:探讨部分CB、TRPV亚型受体参与多不饱和脂肪酸提高大鼠空间学习记忆的过程。将56只初断乳的SD大鼠随机分成7组:二十二碳六烯酸(DHA)组、核桃油组、紫苏油组、红花籽油组、α-亚油酸组、必需脂肪酸(essential fatty acid,EFA)缺乏组和阴性对照组。连续喂养8周后进行Morris水迷宫行为学测试,并通过RT-PCR的方法测定大鼠脑部海马组织中两种大麻素受体亚型(CB1-2)和四种瞬时受体电位香草醛亚型(TRPV1-4)m RNA的表达水平。结果显示,喂食各种油的大鼠在Morris水迷宫实验中的成绩得到明显提高。并且喂食各种油的大鼠脑部海马组织中CB1和TRPV1的m RNA表达量明显提高,TRPV2的m RNA表达量明显降低,对其他受体亚型则没有影响。提示多不饱和脂肪酸能显著提高大鼠的学习记忆能力,其机制与调节CB1,TRPV1和TRPV2的m RNA表达量有关。To explore the mechanism of the rats' spatial learning and memory enhanced by some CB and TRPV subtype receptors involved in polyunsaturated fatty acids. 56 male SD rats (early weaning) were randomly divided into seven groups,DHA group,walnut oil group,perilla seed oil group,safflower oil group,a-linoleic acid group,EFA lack group and negative control group. After 8 weeks of gavage,Ethnology test was done by Morris water maze ,and determined the expression of receptor potential vanillin subtypes (TRPVl -4) mRNA cannabinoid receptor subtypes (CB1 -2) and transient in the hippocampus of rats' brain with the method of RT-PCR. It showed that the score of the rats fed by kinds of oils in the Morris water maze improved significantly and the mRNA expression quantity of CBland TRPVl of hippocampus of rats significantly increased,while TRPV2 reduced very obviously,and there was no influence on other subtype receptors. It proved that polyunsaturated fatty acids could improve the rats' spatial learning and memory significantly,related with the expression quantity of mRNA of CB1 ,TRPVl and TRPV2.
