摘  要:蛹虫草可作为野生冬虫夏草的替代品,虫草素为其重要药用成份,具有较高的药用价值,由于其生产成本较高,迫切需要降低虫草素的生产成本。为了探索高效低成本的虫草素生产工艺条件,本试验对3株蛹虫草优良菌株的生产性能进行了比较,并对4种固态发酵生产虫草素的培养基配方进行了筛选,考察了3种大孔树脂对于虫草素的吸附分离效果,优化了虫草素高效制备的工艺条件。结果表明菌株GIM5.512在子实体和栽培料中的虫草素含量最高,其最适合虫草素生产的固态发酵料配方为:大米35g+营养液55mL。营养液配方为:葡萄糖30g,磷酸二氢钾10g,维生素C10g,维生素B110g,水1000mL。DM130树脂为最适宜吸附分离虫草素的树脂。其对虫草素的吸附率和解吸附率分别可达83.47%和51.97%,当洗脱液乙醇浓度为25%、pH值为5.0时,DM130树脂的解吸附效果最好。精制后样品的虫草素纯度可达85.02%。该研究对于高效生产和制备虫草素具有重要的实际生产意义。Cordyceps militaris is a valuable medicinal fungi. It could be used as a succedaneum of Cordyceps sinensis.Cordycepin is one of the most important pharmaceutical components from C. militaris. However,the cost of codycepin production is very expensive. It is desperately needs to reduce the cost in cordycepin production. In this study,the production traits of three C. militaris strains were compared, and four culture mediums suitable for cordycepin production by solid-state fementation were studied. The processes were optimized for isolation and separation of cordycepin by three kinds of macroporous. Results showed that the strain GIM 5. 512 was the most suitable strain for production of cordycepin. The most suitable culture medium for cordycepin production by solid-state fementation was rice35 g and nutrient solution 55 m L. The nutrient solution fomula was consisted of: glucose 30 g,momopotassium phosphate 10 g,vitamin C 10 g,vitamin B1 10 g,and water 1 000 m L. Macroporous resin DM130 is the best resin suitable for isolation and separation of cordycepin. The optimum conditions for cordycepin desorption is when ethanol concentration is at 20% and p H 5. 0. The adsorption and desorption rate were 83. 47% and 51. 97%,respectively. The purity of cordycepin is 85. 02%. This study was important for lower the cost of cordycepin production in practice.
