摘  要:目的:探讨超滤法去除食源性低聚肽中细菌内毒素的效果。方法:采用凝胶法半定量检测5种食源性低聚肽中细菌内毒素的含量;利用膜材料为聚醚砜、截留分子质量为3000、5000、10000、30000、50000的超滤离心管去除样品溶液中的内毒素,评价不同截留分子质量超滤膜的内毒素清除效果;综合考虑内毒素清除率、超滤速度、生产效率、产品回收率等因素,选定截留分子质量为13000的卷式超滤膜进行工艺放大。结果:截留分子质量为3000、5000、10000的超滤膜可将内毒素控制在50 EU/g以内,截留分子质量为30000和50000的超滤膜可分别将内毒素控制在50-200 EU/g、200-400 EU/g范围内;经截留分子质量为13000的卷式超滤膜超滤后,食源性低聚肽样品中细菌内毒素〈50 EU/g,清除率〉97.5%,产品回收率〉75%。结论:超滤法可有效去除食源性低聚肽中细菌内毒素,并且具有可重复利用、生产成本低等优点,适合用于工业化生产中。Objective: This research aimed at estimating the effect of ultrafiltration technology in eliminating the bacterial endotoxin from food-origin oligopeptides. Methods: Tachypleus Amebocyte Lysate(TAL) assay was used to determine the level of endotoxin in 5 kinds of oligopeptides. To remove the endotoxin from samples, ultrafiltration centrifuge tubes made of polyether sulfone(PES) were used, of which the molecular weight cut-off are 3000, 5000, 10000, 30000 and 50000. Then, based on comprehensive analysis of factors such as clearance rate, speed of ultrafiltration, production efficiency and product recovery rate, can membrane with 13000 molecular weight cut-off was used in amplification process. Results: Ultrafiltration membrane with 3000, 5000 and 10000 molecular weight cut-off can make thecontent of endotoxin below 50 EU/g; the membrane with 30000 and 50000 molecular weight cut-off could make the level of endotoxin in the range of 50-200 EU/g and 200-400 EU/g respectively. Can membrane with 13000 molecular weight cut-off can control endotoxin below 50 EU/g, and with the clearance rate97.5% and product recovery rate75%. Conclusion: Ultrafiltration technology can effectively remove the bacterial endotoxin from food-origin oligopeptides. Besides, the ultrafiltration membrane can be repeated used and the purifi cation cost is low to some extent. Thus, ultrafiltration technology is suitable for industrialized production of food-origin oligopeptides.
