摘 要:微波稳定化处理小麦胚为原料,研究影响超临界CO2萃取小麦胚芽油的因素,并利用有影响贡献的因素优化了超临界CO2萃取小麦胚芽油的工艺.选取小麦胚粉碎颗粒目数、麦胚水分含量、萃取压力、萃取温度、萃取剂流量和萃取时间为试验影响因素,利用Plackett-Burman进行试验设计,筛选影响超临界CO2萃取小麦胚芽油的因素.根据筛选出的因素,利用Box-Behnken进行响应曲面优化水平组合试验.结果显示,因素影响从大到小依次为萃取剂流量>萃取压力和萃取时间>萃取温度>麦胚目数,而3.3%~4.7%样本水分对萃取影响极小;响应面分析试验得到小麦胚芽油萃取率回归方程,方程达到极显著水平,拟和很好.超临界CO2萃取小麦胚芽油最佳工艺参数:麦胚目数为60目、萃取剂流量为30 L/h、萃取压力为25 MPa、萃取时间为141 min和萃取温度40℃.在此条件下小麦胚芽油萃取率达到82.79%.The wheat germ used as raw material was stably treated by microwave irradiation. The wheat germ oil supercritical CO2 extraction factors were studied and the influential contributing factors were used to optimize wheat germ oil supercritical CO2 extraction process. The wheat germ smash particles mesh, wheat germ moisture content, extraction pressure, temperature, flow rate and time were selected for the experimental factors. The factors of super- critical CO: extraction of wheat germ oil were screened by using Plackett - Burman experimental design, which were used for the Box - Behnken response surface optimization level combination trials. The results showed that the impact of factors in decreasing order was extractant flow 〉 extraction pressure and extraction time 〉 extraction temperature 〉 wheat germ mesh, and with water mass ratio from 3.3% to 4.7% samples had minimal impact on the extraction;The regression equation of wheat germ oil extraction rate was obtained through experiment of response surface analysis, which reached a significant level and fitted very well. Optimum process parameters were got by wheat germ oil supercritical CO2 extraction: wheat germ mesh 60 mesh, extractant flow 30 L/h, extraction pressure 25 MPa, extraction time 141min and the extraction temperature 40 ℃. In this condition, the wheat germ oil extraction rate reached 82.79%.
