拟目乌贼生殖腺多糖提取工艺优化及自由基清除能力研究

摘　　要：以拟目乌贼生殖腺为材料,在对料液比、提取时间及提取温度进行单因素实验的基础上,采用Box-Benhken实验设计优化热水浸提法提取多糖工艺,并通过清除超氧阴离子和羟基自由基的能力探究其体外抗氧化活性。结果表明:拟目乌贼生殖腺多糖热水浸提法最佳提取工艺条件为料液比1∶30g/m L,提取温度90℃,提取时间1h,在此条件下拟目乌贼生殖腺多糖得率为0.751%;拟目乌贼生殖腺多糖对超氧阴离子自由基和羟自由基均有一定的清除能力,清除率与多糖浓度呈正相关性;红外图谱显示该多糖主要是β-型糖苷键连接的吡喃型葡聚糖。Using the gonad of Sepia lycidas as raw material,optimization of extraction polysaccharides process was studied by using Box-Behnken design based on the results of single-factor test; antioxidant activity of polysaccharides was evaluated by its superoxide anion and hydroxyl free radical scavenging abilities.The results showed that the optimal extraction parameters were as follows：1：30g/mL for ratio of raw material to water,90℃ for extraction temperature, and 1h for extraction time. Under the optimal conditions, the extraction rate of polysaccharides was 0.751%.The polysaccharides had good abilities to scavenge hydroxyl and superoxide anion free radical.It was indicated to a positive correlation with concentration of polysaccharides. IR spectrum showed that the polysaccharide was pyranoid glucan with 15-glycosidic bond.
