摘  要:为优化梅奇酵母的培养条件,研究不同温度、初始p H值和接种量对梅奇酵母生物量的影响。通过中心复合试验设计,构建3因素(温度、初始p H、接种量)5水平的响应面设计方法,建立梅奇酵母生长的活菌数与3因素的二次回归数学模型。结果表明:3因素对梅奇酵母的生长均有显著影响(P<0.01),影响的顺序为温度>初始p H值>接种量;初始p H值与接种量之间存在显著的交互作用(P<0.01),温度与初始p H值、接种量之间的相互作用对梅奇酵母生长的影响不显著(P>0.05);多元二次回归分析结果显示,温度、初始p H值、接种量与梅奇酵母菌生物量之间的回归模型高度显著(P<0.01)。统计分析确定梅奇酵母的最佳摇瓶培养条件为:温度26.1℃、初始p H 5.6、接种量1.2%。在此条件下,梅奇酵母活菌数的对数预测值为9.3912,验证试验所测浓度为2.56×109CFU/m L,换算为lg对数即为9.408,与模型相比,误差为0.19%。To obtain optimal cultivation conditions for Metschnikowia zizyphicola, an experiment was conducted to explore the effects of different temperature, initial PH and inoculation amount on the maximum biomass of the yeast. The response surface methodology was applied to optimize the main cuhivation factors including temperature, initial PH and inoculation amount, and a quadratic regression equation was established.The results show that the temperature, initial PH and inoculation amount have noticeably significant effects on the biomass of the yeast (P〈0.01), and the order of the significance is: temperature 〉 initial PH 〉 inoculation amount. There are obvious interactions between the initial PH and vaccination concentration (P〈0.01). However, the interaction between the temperature and initial PH, and inoculation amount is not significant (P〉0.05). The multivariate quadratic regression model of the biomass of the yeast and the temperature, initial PH and inoculation amount is highly significant(P〈0.01). The optimal cultivation conditions are that the temperature is 26.1℃, the initial PH is 5.6, and the inoculation amount is 1.2%. Under these conditions, the biomass of the yeast is 2.46×10^9 CFU/ mL, while the biomass from the test is 2.56×10^9CFU/mL. The research lays a foundation for expand cultivation and usage further of Metschnikowia zizyphicola.
