摘 要:运用超声辅助90%乙醇溶液提取芝麻素。在单因素实验的基础上,运用正交实验优化芝麻素超声辅助提取工艺参数。普鲁士蓝法测定芝麻素还原能力,滤纸片-抑菌圈法测定其抗菌活性。结果表明:超声辅助提取的最优条件为超声功率300 W、液固比25∶1、超声温度55℃、超声时间25min,在最优条件下,芝麻素提取率为(3.942±0.085)mg/g;芝麻素具有一定的还原能力,但比相同质量浓度的VC弱得多;当质量浓度大于40 mg/m L时,芝麻素对蜡状芽孢杆菌、金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌均有不同程度的抑制作用,质量浓度大于60 mg/m L时其对4种细菌抑制作用的强弱顺序为蜡状芽孢杆菌>金黄色葡萄球菌>枯草芽孢杆菌>大肠杆菌,而当质量浓度小于100 mg/m L时,芝麻素对真菌黄曲霉无抑制作用。Sesamin was extracted by ultrasound-assisted 90% ethanol solution. On the basis of single factor experiment,the parameters of the ultrasound-assisted extraction of sesamin were optimized by orthogonal experiment. The reducing activity and the antibacterial activity of sesamin were determined by Prussian blue method and filter paper-inhibition zone method,respectively. The results showed that the optimal ultrasound-assisted extraction conditions were obtained as follows: ultrasonic power 300 W,ratio of liquid to solid 25∶ 1,ultrasonic temperature 55 ℃ and ultrasonic time 25 min. Under the optimal conditions,the extraction rate of sesamin was( 3. 942 ± 0. 085) mg / g. Sesamin had certain reducing activity,much weaker than VCof the same mass concentration. When the mass concentration of sesamin was higher than 40 mg / mL,sesamin showed different degrees of inhibitory action on Bacillus cereus,Staphylococcus aureus,Bacillus subtilis and Escherichia coli,and the inhibitory action of sesamin with mass concentration higher than 60 mg / mL on Bacillus cereus was the strongest,followed by Staphylococcus aureus,Bacillus subtilis and Escherichia coli. When the mass concentration of sesamin was less than 100 mg / mL,no obvious inhibitory action of sesamin on Aspergillus flavus was observed.
