摘  要:对从泡菜中分离到的乳酸菌3-9菌株采用固定化技术转化菜油脚料生成共轭亚油酸进行研究。首先对乳酸菌3-9细胞的固定吸附材料及固定化条件进行探讨,确定最适固定化材料为直径1-3 mm的瓷颗粒,细胞吸附固定化时间是1 h,细胞固定率达到75%。将该固定化细胞接入经爪哇毛霉脂肪酶水解后的菜油脚介质转化液中,转化生成的CLA量为(187.91±3.71)μg·m L^-1,是未固定化细胞CLA产量的1.16倍。随后对固定化乳酸菌3-9转化菜油脚生成共轭亚油酸条件进行研究。当以p H8.0的磷酸缓冲液为转化介质基础液,转化时间为17 h、转化温度为40℃、固定化的细胞浓度为0.5 g·m L^-1及摇床转速为120 r·min^-1时,CLA的生成量可达(261.65±3.21)μg·m L^-1,转化率为17.64%。该固定化细胞能连续转化利用16个批次,前10个批次CLA产量均保持在200μg·m L^-1以上,其最高产量可达(283.81±6.02)μg·m L^-1,转化率为19.14%。最后对经1、8和16批次转化后的固定化细胞瓷颗粒进行电镜扫描观察,图片显示经8批次的转化利用后,仍可见大量形态完整的乳酸菌细胞吸咐固定于瓷颗粒中。In this paper, lactic acid bacteria 3-9 screened from pickles was used to convert rapeseed oil sediment into conjugated linoleic acid. Simultaneously, the cells of lactic acid bacteria 3-9 were handled for immobilization. Firstly, adsorption immobilization conditions for lactic acid bacteria 3-9 were investigated and the optimum immobilization conditions were: using 1-3 mm ceramic particles to immobilize the cells for 1 h. The immobilization rate of the cells could reach to 75%. The immobilized cells were placed into rapeseed oil sediment conversion liquid that was hydrolyzed by mucor javanicus lipase and the yield of CLA was 187.91±3.71 μg·m L^-1, which was 1.16 times higher compared to the resting cells. Secondly, the conditions for immobilized lactic acid bacteria 3-9 to convert rapeseed oil sediment into conjugated linoleic acid were studied. When phosphate buffer(p H8.0) was used as the transformation base liquid, under the conditions of 17 h transformation at 40℃, the cell concentration at 0.5 g·m L^-1, and the oscillation speed at 120 r·min^-1, the yield of CLA could reach to 261.65±3.21 μg·m L^-1 and the conversion rate was 17.64%. These immobilized cells can continuously convert 16 batches of rapeseed oil sediment. The yield of CLA was higher than 200 μg·m L^-1 among the 10 batches. The highest yield could reach to 283.81±6.02 μg·m L^-1 and the conversion rate was 19.14%. Lastly, the immobilized cell granules were observed by SEM after conversion of 1, 8, and 16 batches. After 8 batches of conversion and utilization, a large number of intact lactic acid bacteria 3-9 cells absorbed in ceramic particles still could be observed.
