摘  要:为了研究保定、石家庄及周边地区生肉来源的单增李斯特菌株彼此之间的亲缘关系,揭示不同来源、不同时间、不同地域的单增李斯特菌流行变化规律,为该病原菌的检测、防治以及溯源提供依据。对从生鲜肉分离出的129株食源性单增李斯特菌进行RAPD扩增,用NYSYS-PCversion 2.1分析软件采用不加权成对算数平均法(UPAMA)进行聚类分析。采用单一引物(RP1)对129株生肉来源的单增李斯特菌进行RAPD聚类分析,得到了129株单增李斯特菌的DNA指纹图谱,绘制出其亲缘关系的聚类树状图。结果表明:129株生肉来源的单增李斯特菌可分成6个基因聚类,其中第Ⅱ类为保定、石家庄地区的主要流行种群;对单增李斯特菌遗传多态性及其随时间、地域及其菌株来源的变化进行分析,表明该地区生肉来源的单增李斯特菌由于取样时间、地点不同,其基因聚类差异明显,相同时间、相同地点的单增李斯特菌株之间亲缘关系较近。本研究表明四种生肉来源的单增李斯特菌具有广泛的遗传多态性;单增李斯特菌的流行具有明显的时间性、地域性。In order to reveal the variation law of Listeria monocytogenes varying with time and region and to provide the basis of detection ,control and traceability on this pathogen ,the genetic background of L .monocytogenes from raw meat sources in Baoding ,Shijiazhuang and the surrounding areas were investigated .Genetic polymorphism of 129 wild isolates of L . monocytogenes which was isolated from raw pork ,raw chicken ,raw beef and raw mutton , was analyzed by RAPD .Genetic distant coefficient analysis of the different band profiles were carried out by using unweighted pairgroup method analysis (UPGMA) and cluster analysis using statistical data software NTSYSPC version 2 .1 .The oligomer (RP1) as only primer was used in typing 129 L .monocytogenes isolates .The RAPD fingerprint on L .monocytogenes was obtained .The genetic relationship dendrogram among the 129 isolates of L .monocytogenes was established .The results of clustering analysis showed that 129 isolates of L .monocytogenes could be divided into six genetic clusters according to their genetic similarity .Among them ,the cluster II was the main popular species in Baoding and Shijiazhuang .It was also found that there were many differences between each cluster that were related to the food type ,sampling time and region .The isolates which derived from the same sampling time and the same sampling area ,were more closely related genetically .This study suggests that the L .monocytogenes isolates which were derived from four kinds of raw meat are highly diverse . The spread of foodborne L .monocytogenes has characteristics of being regional and temporary .
