摘 要:采用MTT比色法研究α-亚麻酸对HepG2细胞的损伤作用,利用倒置相差显微镜观察HepG2细胞形态,并通过测定ROS水平、SOD活性以及MDA含量来探讨α-亚麻酸对HepG2细胞氧化应激的影响。结果表明:α-亚麻酸在50-250μmol/L浓度范围内呈剂量-时间依赖性方式抑制HepG2细胞的生长增殖,说明α-亚麻酸能够诱导HepG2细胞损伤;α-亚麻酸在200μmol/L浓度下作用24 h后HepG2细胞形态发生明显改变,并在高浓度时HepG2细胞呈典型的凋亡形态学改变;α-亚麻酸作用HepG2细胞后致使细胞大量生成ROS,SOD活性下降,MDA含量升高,这说明α-亚麻酸对HepG2细胞氧化应激产生影响,从而诱导HepG2细胞损伤。The injury of a- linolenic acid on HepG2 cell was investigated by MTr colorimetric method,inverted phase contrast microscope was used to observe morphology of HepG2 cell and the reactive oxygenspecies (ROS) level, superoxide dismutase (SOD) activity and malonadialdehyde (MDA) content weredetermined to discuss the effect of a - linolenic acid on the oxidative stress of HepG2 cell. The resultsshowed that the proliferation of HepG2 cell was inhibited by c~ - linolenie acid in a dosage - dependent(50- 250 μmol/L) and time- dependent manner, which indicated that ol -linolenic acid could inducethe injury of HepG2 cell. Themorphology of HepG2 cell changedafter incubated with 200 μmol/L a -linolenic acid for 24 h and showed typical apoptotic morphology changes in high concentration. Aftertreatment with a-linolenic acid, the contents of ROS and MDA in HepG2 cell increased, while the activi-ty of SOD decreased, indicating that x - linolenic acid affected the oxidative stress of HepG2 cell to in-duce the injury of HepG2 cell.
