摘  要:运用响应面分析研究了用草鱼鱼鳞制备柠檬酸-苹果酸钙的工艺,探讨了柠檬酸-苹果酸有机混酸对草鱼鱼鳞中钙的提取效果.考察了液料比、温度、柠檬酸和苹果酸的比例3个因素对钙提取率的影响.试验结果表明,柠檬酸-苹果酸钙的最佳提取工艺为:液料比为18.5∶1,提取温度为61℃,苹果酸和柠檬酸比例为1.1∶1.在中心组合试验设计建立的回归模型中,失拟项不显著,模型的拟合程度较好.验证试验表明,最高钙提取率为8.56%,与理论预测值接近.本研究为草鱼加工下脚料资源开发和利用提供了理论依据,对实际生产具有指导意义.Calcium citrate-malate is better than other calcium salts as a health supplement due to its better solubility, absorption and safety. The present study was to examine the effect of mixed organic acids citrate and malate on the calcium extraction from grass carp scales using response surface methodology. Meanwhile, the effect of three factors including liquid ratio, temperature and the ratio of citric acid to malic acid on the extraction yield was analyzed. Results showed that the optimized extraction condition was: the ratio of material to liquid of 18.5:1, extraction temperature of 61℃ and the ratio of malic acid to citric acid of 1.1:1. In the regression model , the lack of fitting items was not significant, while the fitting degree was better. The validation test results showed that the highest calcium extraction yield was 8.56% which was close to the theoretical prediction value. By response surface analysis, the order of the effect on the calcium extraction of the three factors was: liquid material ratio, malic acid to citric ratio, and temperature. The effect of the interaction of liquid to material ratio and temperature on the calcium extraction was significant. In the same liquid to material ratio, the extraction yield was increased firstly and then decreased with the increase of temperature. This was because the calcium was more soluble in the form of calcium salts, if the temperature was too high, it would cause the water solubility of calcium salts decreased. Due to the poor thermal stability of citric acid, other chemical reaction would occur leading to extraction rate reduction. Effects of interaction of liquid to material ratio and malic acid to citric acid ratio on the extraction rate of calcium were extremely significant. Under different ratio of liquid to material, the optimal ratio of malic acid to citric acid was different, which may be due to the extraction of calcium was the mixed calcium salt which had not yet been fully defined the chemical structure.
