摘 要:目的建立快速测定葡萄酒中的组胺、尸胺、腐胺、酪胺、色胺、精胺、亚精胺和β-苯乙胺的液相色谱分析方法。方法采用邻苯二甲醛/巯基乙胺混合溶液为衍生化试剂,设置在线自动衍生化进样程序,反相高效液相色谱法测定葡萄酒中的8种生物胺。结果在0.25~10 mg/L范围内,液相色谱的荧光响应值与葡萄酒中生物胺含量呈线性关系,8种生物胺的检出限在20~100μg/L范围之间,RSD〈5%,日间精密度在2.5%~6.3%之间,加标回收率在91.2%~104.1%之间。结论本方法在25 min内能够测定葡萄酒中的8种生物胺,操作简单、结果准确可靠、重现性好。Objective This paper reports a new, simple and rapid method to determine histamine, cadaverine, putrescine, tyramine, tryptamine, spermine, spermidine and β-phenylethylamine in wine without sample pretreatment. Methods Following automatic online derivatization with a mixed solution of cysteamine and o-phthaldialdehyde, eight kinds of biogenic amines in wine were separated on a high performance liquid chromatography instrument with a C18 column. Results The linear range of the proposed method was validated in the range O. 25-10 mg/L. The relative standard deviation was less than 5% , and the intermediate precision ranged from 2.5 to 6. 3%. The recoveries varied from 91.2% to 104.1% , and the detection limits varied from 20 to 100 μg/L for the eight kinds of biogenic amines. Conclusion The method is able to accurately determine the content of eight kinds of biogenic amines in wine with good reproducibility, accurate quantization and without sample pretreatment.
