摘  要:【目的】优化碱蓬多糖的提取工艺,并研究其抗氧化性,为碱蓬多糖的深入开发与利用提供参考。【方法】通过单因素试验考察料液比、Na OH溶液浓度、提取温度、提取时间对碱蓬多糖提取率的影响,并采用响应面分析法优化碱蓬多糖的提取工艺条件;同时采用化学法测定碱蓬多糖对羟基自由基(·OH)、超氧阴离子自由基()和DPPH自由基(DPPH·)的清除作用。【结果】料液比、提取温度和提取时间对碱蓬多糖提取率有极显著影响(P<0.01),Na OH溶液浓度及其与提取时间、料液比与提取时间的交互作用对碱蓬多糖提取率有显著影响(P<0.05)。响应面法优化得到碱蓬多糖的最佳提取工艺为:以0.17 mol/L Na OH溶液为提取溶剂,在料液比1∶42.5、提取温度83.5℃的条件下提取3.4 h,多糖提取率为8.00%,与理论值8.02%非常接近。碱蓬多糖对·OH、和DPPH·均有明显的清除作用,且清除率随多糖质量浓度的增加而增大,最高清除率分别为89.15%、80.87%和88.12%。【结论】采用响应面分析法优化碱蓬多糖提取工艺具有可行性,可提高多糖提取率;碱蓬多糖具有明显的抗氧化作用。[Objective]The present experiment was conducted to optimize extraction process of polysaccharide from Suaeda sa/sa and study its antioxidation ativity, in order to provide references for development and utilization of S. salsa polysaccharide. [Method ]The effects of solid-liquid ratio, NaOH concentration, temperature and time on extraction rate of S. sa/sa polysaccharides were investigated based on single factor test. Then the extraction process of S. salsa polysaccha- rides was optimized by response surface analysis (RSA). Meanwhile, the scavenging effect of S. salsa polysaccharides on hydroxyl radical (·OH), superoxide radical(O2^-)and DPPH radical(DPPH. )were analyzed through chemical assay. [Resuit]The material-liquid ratio, extraction temperature and extraction time have extremely significant effects on extraction rate of S. salsa polysaccharides (P〈0.01). The NaOH concentration and its interaction with extraction time, and interaction between solid-liquid ratio and extraction time had significant effects on extraction rate of S. sa/sa polysaccharides (P〈 0.05). The extraction process conditions optimized by response surface methodology were as follow: 0.17 mol/L NaOH concentration as extraction solvent, solid-liquid ratio of 1:42.5, extraction temperature of 83.5 ℃, extraction time of 3.4 h. Under the above optimal process conditions, the extraction rate of polysaccharide was 8.00%, which approached to theo- retical value 8.02%. The scavenging effect of S. salsa polysaccharide on · OH, O2^-: and DPPH. were obvious and effective, which increased with the increasing of polysaccharide concentrations, and the hightest scavenging rate of · OH, O2^- and DPPH. was 89.15%, 80.87% and 88.12%, respectively. [Conclusion]The response surface methodology is a workable method to optimize extrcation process of S. sa/sa polysaccharides, which improve effectively extraction rate of polysaccha- rides. Furthermore, S. sa/sa polysaccharides show obvious antioxidation effect.
