摘  要:用木瓜蛋白酶制备核桃仁蛋白酶水解物,通过细胞增殖分析试验,研究其对人乳腺癌细胞(MCF-7)增殖的影响,结果显示:水解物具有明显的抑制细胞生长作用,IC50值为1.62mg/m L。用超滤方法将核桃仁蛋白酶水解物,分为10~5k Da、5~3k Da和〈3k Da3个组分,研究不同分子量范围的组分对MCF-7细胞增殖影响的结果表明,MW〈3k Da的低分子量组分具有较强的增殖抑制作用。对3个不同组分进行氨基酸含量分析,结果表明:具有较强抑制癌细胞生长作用的低分子量组分中,疏水性氨基酸及中性极性氨基酸含量较高,酸性极性氨基酸含量少于其他组分。Walnut protein hydrolysate was prepared with papain, and its effect on proliferation of human breast cancer cell ( MCF-7 ) was determined by MTF assay. Results showed that the walnut protein hydrolyzed inhibited proliferation of the cells significantly ( IC50 value was 1.62mg/mL) . Walnut protein hydrolysate was separated to three fraction of 10 -5kDa, 5 -3kDa and 〈 3kDa by ultrafihration. The effect of the bydrolysates with different molecular weight on MCF-7 cells proliferation was investigated. Results showed that MW 〈 3 000kDa fraction with lower molecular weight showed the higher inhibitory activity among the three fractions. The amino acid compositions of the three fractions with different molecular weight were analyzed. The results showed that content of hydrophobic and neutral polar amino acids in MW 〈 3 000 kDa fraction was higher than the other fractions, while the content of acidic polar amino acid in MW 〈 3 000kDa fraction was lower than the other fractions.
