摘  要:选取3种沙门氏菌快速检测法,以国标法为参考,比较几类方法检测沙门氏菌的特异性、灵敏度和抗干扰性。并研究添加成分和食品基质对检测效果的影响。结果表明,MiniVIDAS法和环介导等温扩增法(Loop-mediated isothermal amplification,简称LAMP)的特异性、灵敏度和抗干扰性较好,测试片法和国标法相对较差。国标法和Mini-VIDAS法的检测结果受添加成分影响较大,测试片法和LAMP法不受其影响。另外,添加成分影响较低浓度沙门氏菌(105 cfu/m L)的检出。4种检测方法受添加成分的影响程度基本符合防腐剂>凉拌菜调味料≥十三香调味料。当食品基质中微生物污染背景较高且沙门氏菌污染剂量较低时,国标法和测试片法的检测结果受食品基质干扰较大,Mini-VIDAS法和LAMP法效果较好。综上所述,MiniVIDAS法和LAMP法是较理想的沙门氏菌快速检测方法。但是国标法仍是目前较通用且准确的方法,操作时需考虑食品基质本身的性质,如微生物污染背景、添加成分等,以提高沙门氏菌检出率。The examination effect of Salmonella with rapid method and GB method were compared in the aspects of specificity, sensitivity and interference. Effects of addictives and food matrix on the detection results were also studied. Experimental results indicated that the methods of mini-VIDAS and Loopmediated isothermal amplification(LAMP) have higher specificity, sensitivity and interference than the methods of testing slip and GB. The tested addictives have no impact on the detection results of Salmonella with the methods of testing slip and LAMP, but have an impact on the results when using the methods of mini-VIDAS and GB. When the number of Salmonella was lower(about 105 cfu/m L), the detection results were intensivily interferred with the addictives. The negative effect of three addictives on Salmonella detection results followed the order: preservativesvegetable salad condiment ≥thirteen-spices. When there are high bacterial background and small infective dose of Salmonella in food matrix, the detection results with the methods of testing slip and GB were intensivily interferred. However, using the methods of miniVIDAS and LAMP, food matrix have no effect on the detection results. Thus, the rapid and effective methods of mini-VIDAS and LAMP are the better one to detect Salmonella in food. But GB method is still a popular and accurate method. Additionally, testing personal should pay more attention on the characteristics of food matrix, such as bacterial number and food addictives.
